Human evolutionary history revealed by variation in African genomes

Genetic variation forms the basic material that allows evolutionary change over time. Finding, describing and utilizing genetic variation to understand human history has long been a focus of evolutionary biology and genetics and the last few years genomic revolution has given us the means to produce immense population-genetic data, which now make it possible to provide novel insights into human evolutionary history.In recent studies of worldwide genomic variation, the Khoe-San are constantly identified as the most divergent group of modern humans. Since the earliest diversification of modern humans involves the ancestors of Khoe-San, the time immediately prior to this event corresponds to the critical period for the emergence of anatomically modern humans.I will characterize complete genomic variation in African groups including the Khoe-San by sequencing genomes of more than 100 individuals, which will dramatically expand the catalog of known human genetic variation. I will use population genetic theory and computational approaches to infer the evolutionary history of African groups. Since the earliest diversification of modern humans involves the ancestors of Khoe-San, I can use the information in these genomes to reveal the process by which modern humans emerged, including the genetic changes that were involved in transforming archaic humans to modern humans. My research program has the potential to answer the ultimate question - what makes us human?
