Nuclear interactions in the fungal heterokaryon
Abstract [en]
Heterokaryosis is a genetic system in which genetically different nuclei coexist in the same cytoplasm. It is widespread among eukaryotes, and is particularly common in the fungal kingdom. Knowledge about the molecular interactions of nuclei in heterokaryons is needed to understand the broader evolutionary and ecological implications of heterokaryosis. Specifically, little is known about how genetically divergent nuclei function in the common cytoplasm; if and how they maintain and diverge functions over evolutionary time. Here, I propose a project to generate transcriptome and methylome data from homokaryotic and heterokaryotic samples of Neurospora, from different developmental stages. Within this genus, a recent shift from homokaryosis to heterokaryosis has taken place, and it is thereby suitable for dissecting gene expression and genomic changes in early stages of heterokaryosis. With this data in hand, we will be able to investigate: i) Phenotypic novelty of heterokaryosis in terms of gene expression, ii) Patterns of coregulation of genes between nuclei, iii) Evolutionary convergence in division of labor between nuclei.  This project will increase our understanding of heterokaryosis, and thereby of fundamental aspects of life history evolution in nature. More generally, this project will broaden concepts of genetic individuality and provide important information about multilevel selection, which occurs both above and below the level of the individual.
